Why is this movie looks black?

Why this movie looks black like a small black cloud is somewhere in the center this is the fist time I see somethink like this

Yes, that dark cloud about twice as large as a thumb is a puzzlement. :)

No its not that its in the center of movie when i go focus up or down its like a cloud and its at the same position

I believe I've seen what you are referring to; I assume it is a shadow (probably from the microscope) due to the way the samples are lit.

I dont think its a shadow it changes shape as u focus up or down

Can someone answer us what it is? I mean from the program? I already have noticed it as a track even if it is not.
